Ce nouvel outil ouvre des pistes pour \u00e9tudier des maladies comme Alzheimer.<\/em><\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong><em>Le r\u00f4le essentiel des cellules immunitaires<\/em><\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Chaque organe de notre corps est surveill\u00e9 par des cellules immunitaires sp\u00e9cialis\u00e9es, les macrophages. Elles vivent longtemps, \u00e9liminent ce qui est ab\u00eem\u00e9 et aident l\u2019organe \u00e0 rester en bonne sant\u00e9. Le foie, le c\u0153ur et les reins ont chacun les leurs. Dans le cerveau, on les appelle la microglie.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Ces cellules proviennent de cellules m\u00e8res qui apparaissent tr\u00e8s t\u00f4t, dans l\u2019embryon. Chez l\u2019humain, cette \u00e9tape est presque impossible \u00e0 observer, car les tissus embryonnaires sont tr\u00e8s difficiles \u00e0 \u00e9tudier. Les \u00e9quipes de Renzo Mancuso et de Bart De Strooper l\u2019ont donc recr\u00e9\u00e9e en laboratoire, \u00e0 partir de cellules souches humaines.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Pour v\u00e9rifier que ces cellules se comportent comme les vraies cellules m\u00e8res, les chercheurs les ont transplant\u00e9es dans un organisme vivant. Elles ont rejoint plusieurs organes et s\u2019y sont adapt\u00e9es : dans le cerveau, elles sont devenues de la microglie, et dans le foie, le c\u0153ur et les reins, des macrophages propres \u00e0 chacun de ces organes. Elles ont m\u00eame adopt\u00e9 le fonctionnement des macrophages humains de chaque organe. C\u2019est donc le milieu o\u00f9 elles arrivent qui leur indique quel r\u00f4le jouer.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u00ab Ce qui est passionnant ici, c\u2019est que nous pouvons d\u00e9sormais produire des cellules humaines qui non seulement ressemblent \u00e0 celles du d\u00e9but du d\u00e9veloppement, mais qui se comportent aussi comme elles \u00bb, explique Renzo Mancuso.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong><em>Nouvelle strat\u00e9gie pour la maladie d\u2019Alzheimer et les maladies apparent\u00e9es<\/em><\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Dans la maladie d\u2019Alzheimer, certaines cellules immunitaires du cerveau semblent ne plus remplir correctement leur r\u00f4le de surveillance et d\u2019entretien. Pouvoir fabriquer ces cellules \u00e0 partir de cellules de d\u00e9part permettrait d\u2019\u00e9tudier ce qui se passe quand elles se d\u00e9r\u00e8glent, et de tester si de nouvelles cellules pourraient un jour prendre le relais.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u00ab Disposer d\u2019un syst\u00e8me qui nous permet d\u2019\u00e9tudier comment ces cellules se d\u00e9veloppent, s\u2019adaptent et comment elles pourraient \u00e9ventuellement \u00eatre remplac\u00e9es nous offre une nouvelle fa\u00e7on d\u2019explorer leur r\u00f4le dans la maladie \u00bb, souligne Bart De Strooper.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u00c0 plus long terme, les chercheurs veulent explorer si des cellules immunitaires ab\u00eem\u00e9es ou d\u00e9faillantes pourraient un jour \u00eatre remplac\u00e9es, notamment dans les maladies neurod\u00e9g\u00e9n\u00e9ratives comme Alzheimer ou la d\u00e9mence frontotemporale.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u00ab Chaque avanc\u00e9e dans la compr\u00e9hension de ces cellules rapproche la recherche de nouvelles r\u00e9ponses pour les personnes touch\u00e9es par la maladie.
